چکیده

The study examined the response of MG63 to Enamel Matrix Derivative (EMD) on the coarse-grit-blasted and acid-etched surface disks. We characterized associated changes in proliferation and differentiation related intracelluar signaling molecular including mitogen activitied protein kibase. The major groups of MAPKs (ERK1/2, cJUN-terminal kinases, JNK and p38-MAPK) have been characterized in previous experiments. We will check which pathways stimulate the proliferation and differentiation of MG63 on the coarse-grit-blasted and acid-etched surface disks by the western blot. EMD regulated the proliferation and differentiation of MG63 on the coarse-grit-blasted and acid-etched surface disks. EMD inhibit the cell proliferation of MG63, increase the differentiation of MG63 and production of osteocalcin and TGF-β1. The signaling pathway induced by END from ERK1/2 JNK and p38 were detected. EMD has been successfully used to affect the proliferation and differentiation of osteoblast on the surface of implants. EMD can accelerate the regeneration of soft and hard tissue in the peri-implantitis disease. EMD can be used in the pero-implantitis disease to regenerate the soft and

Iranian crack induces hepatic injury through mitogen-activated protein kinase pathway in the liver of Wistar rat

Objective(s): Iranian crack (IC) is a heroin-based substance manifesting various pathologic side effects. Herein, we aimed to investigate the mechanism of IC-induced liver injuries in Wistar rats. Materials and Methods: Twenty male Wistar rats were randomly divided into two groups: control, and IC (0.9 mg/kg/day/IP, for 30 days). Mitochondrial reactive oxygen species (ROS) production was measur...

Osthole-mediated cell differentiation through bone morphogenetic protein-2/p38 and extracellular signal-regulated kinase 1/2 pathway in human osteoblast cells.

The survival of osteoblast cells is one of the determinants of the development of osteoporosis in patients. Osthole (7-methoxy-8-isopentenoxycoumarin) is a coumarin derivative present in many medicinal plants. By means of alkaline phosphatase (ALP) activity, osteocalcin, osteopontin, and type I collagen, enzyme-linked immunosorbent assay, we have shown that osthole exhibits a significant induct...

Regulation of alkaline phosphatase activity by p38 MAP kinase in response to activation of Gi protein-coupled receptors by epinephrine in osteoblast-like cells.

The signaling mechanisms responsible for the regulation of alkaline phosphatase (ALP) activity by exogenous factors in osteoblast-like cells remain poorly understood. Among various agents, epinephrine was recently found to increase ALP activity in differentiating MC3T3-E1 cells by stimulating alpha1 adrenergic receptors coupled to Gi proteins. In the present study, we investigated the role of b...

SP600125 reduces lipopolysaccharide-induced apoptosis and restores the early-stage differentiation of osteoblasts inhibited by LPS through the MAPK pathway in MC3T3-E1 cells.

Bone degradation is a serious complication of chronic inflammatory diseases such as septic arthritis, osteomyelitis, and infected orthopedic implant failure. Effective therapeutic treatments for bacteria-caused bone destruction are limited. In a previous study, we found that lipopolysaccharide (LPS) induced osteoblast apoptosis and inhibited early and late-stage differentiation of osteoblasts v...

Erk is essential for growth, differentiation, integrin expression, and cell function in human osteoblastic cells.

Extracellular signal-regulated kinases (Erks), members of the mitogen-activated protein kinase superfamily, play an important role in cell proliferation and differentiation. In this study we employed a dominant negative approach to determine the role of Erks in the regulation of human osteoblastic cell function. Human osteoblastic cells were transduced with a pseudotyped retrovirus encoding eit...
